What is avuncular testing?

Avuncular testing is a DNA test used to determine if a man or woman is the biological aunt or uncle of the child in question.

On average, an aunt or uncle shares about 25% of their DNA with their niece or nephew, and the process involves comparing 24 genetic markers of the child with those of the potential aunt or uncle to identify any shared genetic material. Based on the number of shared markers, a percentage probability is calculated to indicate a biological relationship.

When is avuncular testing needed?

Avuncular testing is completed for a wide variety of reasons, providing valuable peace of mind, and is often used for legal purposes when direct paternity or maternity testing is not feasible. Reasons for avuncular testing include:

  • Peace of mind reasons – There may be instances where individuals look to confirm the biological connection to a niece/nephew or aunt/uncle for personal reassurance or to understand their family history. This clarity can be especially meaningful where the suspected parent is not available.
  • Missing parent scenario – The potential parent may be unavailable for DNA testing due to various reasons. This could include situations where they are deceased, unwilling to participate, unreachable or absent.
  • Immigration cases Family relationships can be important requirements for immigration applications. Where direct maternity or paternity cannot be established, avuncular testing can provide evidence to establish the biological relationship between a petitioner and beneficiary.
  • Legal purposes
    • Child custody – Avuncular testing can provide important evidence to dispute or support claims of biological paternity or maternity, influencing decisions regarding parental rights, responsibilities and guardianship in the best interest of the child.
    • Inheritance disputes/probate – In cases where there are disputes over an estate or inheritance, avuncular testing can be used to establish inheritance rights. If an individual’s biological relationship to a deceased individual needs to be proven, an aunt or uncle’s DNA can help to prove or disprove the relationship for legal claims.

Key considerations

There are several factors to take into consideration when completing an avuncular test:

Probability – While paternity testing typically provides a highly conclusive result, avuncular testing is based on probability. This is because the inheritance pattern from an aunt/uncle to a niece/nephew is more indirect.

To improve the likelihood of gaining a conclusive result, we highly recommend that the child’s biological mother participates in the test wherever possible. This allows our team of scientists to exclude the genetic material that the child inherited from their mother and focus on paternal genetic markers, providing a clearer picture of the potential relationship and therefore a more definitive probability. Similarly, if the test is to establish a maternal avuncular relationship, the known father’s participation would enhance the results.

Other potential relationships – Avuncular testing is not suitable in cases where the potential aunt or uncle may be related to the child in another way, such as siblings.

Siblingship – Additionally, the test is not suitable if the potential aunt or uncle are only half siblings to the mother or father of the child in question, as this would not produce a reliable result.

The NorthGene™  testing process

Completing a NorthGene™  avuncular test is quick and easy, and can be done in the comfort of your own home:

  1. Order your at-home test kit online.
  2. Once you receive your kit, populate all sections of the sample consent form – date and sign where indicated.
  3. For each person completing the test, fill out all required details on the sample collection envelopes. There will be one envelope per person.
  4. To collect the DNA sample, simply rub a test swab on the inside of the individual’s cheek for 20 seconds. This collects a small sample of cells which is needed to run the test.
  5. Put the swab into the packet and then take out the second. Two sterilised test swabs must be completed for each person participating.
  6. Put the swab packet into the relevant envelope and seal securely.
  7. Once all samples are taken, put the form and all sealed sample collection envelopes in the prepaid tamper-proof bag addressed to NorthGene™ and return via your nearest post box or post office.
  8. Results will be sent to you via email within 1-2 working days upon receipt at our Deeside-based laboratory.
