We know doing an DNA test can be scary. The fear of receiving your results, if a mistake was made during processing, or even doing the test in the first place. So, let’s take a step back from the stress and go through exactly how we at NorthGene™ process your samples to ensure that you get reliable, accurate and fast results.
Pre-testing prep
When your samples arrive in our Deeside based laboratory, before beginning any lab work, we check and confirm all necessary consent has been provided from everyone participating.
Next, your samples are assigned a unique sample identification number, also known as an S number, and barcode to ensure all your samples can be tracked as belonging to you through the entire process on our system, as well as ensuring each sample is attached to their respective consent forms. Once all samples are assigned an S number and barcode, testing can begin.
Step 1: Extracting DNA
To extract DNA from the sample swabs, we cut the tip of the swab off and place it into a corresponding sample test tube. A swab solution is then added into the test tube.
Next, the tube is then placed into a vortex machine and heated to release the DNA into the solution – Imagine a very fast, very small and very hot private use washing machine!
After heating, the tubes are cooled, ready for the next stage of the process.
Step 2: Hot plates
Whilst your samples are cooling, a plate map is created, which tells us exactly where your S number will be placed on a well plate, which can hold up to 96 different samples. Your DNA is then transferred into the well plate, which already contains the master mix – this process includes a secondary witness to ensure complete accuracy.
Your DNA is then once again heated in a cycle to split it into two strands and create copies – this process is called amplification.
Step 3: Electrophoresis
We then transfer your amplified DNA to a genetic analyser to undergo a process called capillary electrophoresis.
Now, imagine a barcode – In this process, we call the individual lines in that barcode a “marker”, this machine reads the individual DNA markers by measuring how long or how short the strands of DNA are and counting how many times they repeat.
These are known as STRs. The number of times it repeats is how many times the markers show up in the “complete barcode”.
Step 4: Analysis

When the machine finishes running, we review the data to make sure all the controls have passed. We then compare the DNA barcode results for each person involved in the case.
Typically, this includes the mother, potential father, and the child. Each child inherits two markers, one from the mother and one from the father.
To determine paternity, we look at the number of these shared markers and the likelihood of the child inheriting these from the potential father
For paternity to be confirmed that is potential father IS the father, the potential father’s markers must match the child.
Numerically, that means the results must show that the child and father must share a 99.99%+ match, otherwise the results are considered inconclusive – meaning anything below 99.99% makes them NOT the father!
Step 5: The final report
When data analysis is complete, we write the report with the concluding results. This is second checked by another member of the team before being sent to the email addresses on the consent form.
At no point does your DNA leave our Deeside based lab, everything is carried out in house. This ensures your active-test safety. After completing the analysis your DNA is then disposed of and your data purged, ensuring your post-test safety!
